Core Facility Confocal Microscopy

Our institute is one of the nodes in a core facility for confocal microscopy financed by Helse Sør-Øst. The two other nodes are located at Montebello and Gaustad.

The Confocal Core Facility Confocal Microscopy Ullevål holds three confocal microscopes and two whole cell fluorescence setups as well as software for image enhancement.

Equipment:

Zeiss LSM 710 / Axio Observer
Inverted confocal microscope used for studying protein localization and colocalization using traditional antibody labelling as well as Duolink. The microscope is also set up for FRET.
Laser lines: 458-488-514-543-633 nm.

Zeiss Live 7 / Axio Observer
Inverted confocal microscope used for studying Ca2+ dynamics in live cells. The Live 7 provides high speed scanning and gives acquisition rates of 1000 images of 512 x 512 pixels in less than 10 seconds. The microscope is set up with a perfusion system (37°C), patch clamp equipment (Axon Axopatch 200B) and field stimulation. Flash photolysis equipment is a also available.
Laser lines: 488-532 nm.

Zeiss LSM 510 / Axiovert 100
Inverted confocal microscope used for studying Ca2+ dynamics in live cells and protein localisation/colocalization. Also equiped with perfusion system (37°C), electrophysiology (Axon Axoclamp 2B) and field stimulation.
Laser lines: 488-543-633 nm.

Zeiss Axiovert 200 / Cairn Optoscan
Inverted widefield microscope used for studying Ca2+ dynamics in live cells. Whole cell fluorescence (for example Fluo-4) can be measured using fluorescence system from Cairn Research (Optosource/Optoscan/1 detection channel). Also equiped with perfusion system (37°C), electrophysiology (Axon Axoclamp 2B) and field stimulation.

Zeiss Axio Observer / PTI
Inverted widefield microscope used for studying Ca2+ dynamics in live cells. Whole cell ratiometric fluorescence (for example Fluo-4, Fura-2, SBFI) can be measured using fluorescence system from Photon Technology International (RatioMaster/2 detection channels). Also equiped with perfusion system (37°C), electrophysiology (Axon Axoclamp 2A) and field stimulation.

Huygens deconvolution software from Scientific Volume Imaging
We hold a licence for use of deconvolution software from SVI which improves image quality by reversing optical distortion in the image.
